Description These mice contain a mutation designed to replace gene
function of endogenous mouse Apoe with the human apoE4
isoform. They also contain a mutation designed to replace
gene function of endogenous mouse Ldlr with human LDLR. Mice
homozygous for Apoe4 and heterozygous for Ldlrh
(Apoe4/4Ldlrh/+) are viable and fertile. The half-life of
Ldlr mRNA is elevated in the liver due to a more stable
3'UTR. Apoe4/4Ldlrh/+ exhibit severe atherosclerosis and a
lower plasma lipid level (presumably due to increased Ldlr
metabolism in the liver). These mice may be useful to
identify the roles human lipoprotein metabolism related
genes in diabetic dyslipidemia and vascular complications.
